Hypoglycemic Activity of Hypholomine B— A Polyphenol from Phellinus igniarius, in Diabetic Mice

Abstract
This study first assessed the in vivo hypoglycemic effect of the styrylpyrone hypholomine B (HB) in diabetic ICR mice. HB was produced by large-scale fermentation. Diabetes was induced with high-fat diet plus streptozotocin. Mice were assigned to normal, model, metformin (100 mg/kg), and HB 10 or 20 mg/kg groups for 35 days of intragastric dosing. Fasting blood glucose (FBG) and oral glucose tolerance (OGTT) were monitored. FBG fell dose-dependently in HB-treated mice; after 35 days both HB doses decreased FBG by -7.0 mmol/L, outperforming metformin (–5.1 mmol/L). OGTT confirmed improved glucose tolerance. Thus, hypholomine B is a potent natural anti-diabetic lead compound.
