mRNA-Based Anticancer Immunotherapy: Molecular Design and Delivery Strategies with Melanoma as a Model

Abstract
mRNA-based anticancer immunotherapy has become a promising cancer treatment strategy, especially after the success of mRNA vaccine technology. mRNA therapeutics can trigger strong immune responses with the expression of tumor-specific antigens and immunomodulatory proteins. This review outlines the molecular design principles of mRNA constructs, including optimization approaches. In addition, current delivery strategies, particularly lipid nanoparticles, are discussed as well. Focused on the high tumor mutation load and immunogenicity of melanoma, it is an ideal model for mRNA-based immunotherapy. Despite the advances, several challenges remain, especially those related to delivery efficiency and immunosuppressive tumor microenvironment. In the future, the strategies of integrating and optimizing the delivery systems, personalizing neoantigen design and combined therapy are expected to promote the clinical application of mRNA-based cancer therapeutics.
