The Effect of Sucrose Induction on Lipid Droplet Hypertrophy and Body Mass Index Increase in Danio rerio

Sucrose is a disaccharide composed of fructose and glucose, is commonly found in various foods and beverages. Excessive sucrose consumption can trigger lipogenesis, leading to obesity, a condition characterized by excessive fat accumulation. This study used an in vivo experimental design with a control group post-test, involving 12-week-old adult zebra fish (D. rerio). Four groups were included: control group (GO) and three sucrose induction treatments: G1 (1%), G2 (2%), and G3 (4%). Each group had 30 fish, induced with sucrose for 28 days. Sacrifice was done by immersing the fish in ice water for 10 minutes. Adipocyte tissue was collected from the abdominal area for histopathological preparation, and BMI was calculated using the D. rerio formula. Lipid droplet hypertrophy was measured using ImageJ software. Data were analyzed with one-way ANOVA (p < 0.05). Sucrose induction increased BMI in Gl, G2, and G3 groups to 0.05±0.011, 0.06±0.007, and 0.08±0.007, respectively, compared to G0 (0.03±0.009). Lipid droplet hypertrophy was higher in treated groups (6.17±0.75, 8.67±0.81, 13.5±2.88) compared to G0 (1.50±1.04). These findings suggest sucrose increases obesity risk in D. rerio. Further studies should explore natural remedies to mitigate sucrose-induced obesity.
