Identification and Initial Testing of Anticancer Compounds from Marine Sponge Extract Clathria sp. From Padamarang Island, Kolaka Regency

A study has been conducted on the identification and initial testing of anticancer compounds from the sponge extract of Clathria sp. from the waters of Padamarang Island, Kolaka Regency. This study aims to identify compounds and conduct toxicity tests of the sponge extract of Clathria sp. Identification was carried out by maceration using methanol solvent, then the methanol extract was partitioned using n- hexane, ethyl acetate and dichloromethane solvents. The partition results were then tested for toxicity using the Brine Shrimp Lethality Test (BSLT) method. Identification of compounds from highly toxic fractions was carried out using a Liquid Chromatography-High Resolution Mass Spectrometer (LC-HRMS) tool. The maceration results obtained a dark yellow and oily methanol extract. The partition results obtained extracts of n-hexane (1.14 g), ethyl acetate (1.81 g), dichloromethane (0.51 g) and methanol/water (1.06 g). The results of the toxicity test showed that the dichloromethane fraction had the most toxic properties with an LC50 value of 1.041 g/mL. LC-HRMS analysis indicated that the dichloromethane fraction contained the compounds Zafaral, Cespitaenin D, and Vibralactone Z4 as well as possibly one new compound. The results of the LC-HRMS analysis of the dichloromethane fraction likely contained 4 compound and 1 new compound that has not been found in the literature.
