This study aims to evaluate the effect of the combination of Ringer’s lactate and Ringer’s acetate diluents supplemented with egg yolk and skim milk as cryoprotectants on abnormality and plasma membrane integrity of Gaga chicken spermatozoa post-cryopreservation. The study design used a Completely Randomised Design (CRD) with four treatments and ten replicates, including Ringer’s lactate + 10% egg yolk (P1), Ringer’s acetate + 10% egg yolk (P2), Ringer’s lactate + 6% skim milk (P3), and Ringer’s acetate + 6% skim milk (P4). The research procedure included semen collection, semen dilution, pre-freezing and freezing of semen, and thawing. The variables observed were the percentage of abnormality and plasma membrane integrity of spermatozoa before and after freezing– thawing. Data were analysed using analysis of variance (ANOVA). Based on the results of this study, the use of Ringer’s lactate and Ringer’s acetate diluents combined with 10% egg yolk or 6% skim milk as cryoprotectants did not result in differences in sperm abnormality and plasma membrane integrity in Gaga chickens during the freezing and thawing process.
Key words: Gaga chicken / cryopreservation / abnormality / plasma membrane integrity / Ringer’s lactate / Ringer’s acetate / egg yolk / skim milk
